GENETIC IMPROVEMENT OF SHEEP BREEDS IN BULGARIA THROUGH APPLICATION OF INNOVATIVE PRODUCT FOR OPTIMIZATION OF BIOTECHNOLOGY FOR IN VITRO STORAGE OF MALE GAMETES

Abstract: Studies have been conducted on the protection mechanism of selected by chromatography seminal plasma proteins on the molecular changes of the plasma membrane (PM), motility and survival rate of ram sperm in order to develop an effective biotechnology for artificial insemination (IS). By fixed three factor interaction model for the analysis of main factors - temperature (T), storage (S) and protein (P) has been found that the proteins in fractions containing higher molecular weight - 100 kDA (fractions 1-3) showed better protection on sperm motility during storage at temperature - 150C, while proteins from fraction 5 and 6 (with molecular weights of proteins 10-50 kDA), protects better sperm motility in the temperature regime of storage - 40C. The results of these studies will help overcome the obstacles on the long-term storage of gametes ram.
